Key Points

  • Oncolytics Biotech has received written FDA feedback on the potential pivotal Part B expansion of its ongoing REO 033 colorectal cancer study.
  • The proposed pathway could use objective response rate and duration of response to support accelerated approval, with progression-free survival potentially supporting full approval.
  • The Simply Wall St community narrative estimates fair value at around $3.50 per share, although its opening valuation estimate is approximately $3.25, highlighting both potential upside and financing risk.

FDA feedback gives REO 033 a clearer path toward pivotal development

Oncolytics Biotech (NASDAQ:ONCY) has received written feedback from the US Food and Drug Administration (FDA) regarding the potential expansion of its ongoing REO 033 study into a pivotal Part B.

REO 033 is evaluating pelareorep alongside FOLFIRI and bevacizumab against FOLFIRI and bevacizumab alone in patients with second-line RAS-mutant, microsatellite stable metastatic colorectal cancer.

The company had asked the FDA whether the current randomized Part A could support a pivotal expansion and whether a subsequent Part B could potentially form the basis for accelerated approval.

The FDA's written response provided what Oncolytics describes as alignment around the proposed development strategy. The regulator also suggested an End-of-Phase meeting to finalize important elements of the registration program.

As a result, Oncolytics plans to skip its previously scheduled Type D meeting and instead pursue an End-of-Phase meeting as the program advances.

Pelareorep could move from clinical testing toward registration

The significance of the feedback is that Oncolytics now has a clearer framework for moving REO 033 toward a potentially registrational study.

The proposed approach could allow objective response rate and duration of response to support an accelerated approval application, while progression-free survival could provide the basis for full approval.

That does not mean approval is assured. The ongoing Part A still needs to generate supportive clinical results, and the company expects those initial data to inform the decision to launch Part B.

Part A includes 60 patients and is intended to evaluate the clinical activity of pelareorep while providing the evidence needed to determine whether the pivotal expansion should proceed.

Oncolytics says it plans to begin Part B start-up activities as soon as positive interim Part A data become available. This could help reduce the time between generating supportive evidence and starting the next stage of development.

The strategy builds on earlier data from REO 022, where pelareorep-containing combinations produced encouraging signals across response rate, duration of response, progression-free survival and overall survival. Pelareorep has also received FDA Fast Track designation in this patient population.

Why this matters for investors

The FDA feedback potentially removes an important layer of regulatory uncertainty around Oncolytics Biotech's colorectal cancer program.

For a clinical-stage biotechnology company, a clearer registration pathway can improve the potential value of positive clinical data. If REO 033 Part A delivers supportive results, the company could potentially move directly into a pivotal Part B rather than redesigning its development strategy from scratch.

However, the announcement does not eliminate the main risks. The regulatory pathway remains conditional on clinical results, while the company will need sufficient capital to fund further development.

The investment case therefore still depends on three factors: whether pelareorep can reproduce its earlier efficacy signals in a randomized setting, whether the FDA ultimately agrees on the final registration program, and whether Oncolytics can finance development without excessive shareholder dilution.

Most followed narrative

The most followed Simply Wall St community narrative presents Oncolytics as a speculative biotechnology opportunity where the market may be placing greater emphasis on financing risk than on the potential value of pelareorep.

The narrative describes pelareorep as an immunotherapy designed to make otherwise "cold" tumors more visible to the immune system and highlights encouraging early data across colorectal, anal and pancreatic cancers.

It also points to the experience of Oncolytics' new management team, including CEO Jared Kelly, who the company says has led more than 50 transactions, including Ambrx Biopharma's approximately $2 billion sale to Johnson & Johnson.

Valuation is a major part of the narrative. It estimates fair value at approximately $3.50 per share in its concluding valuation scenario, while its opening key takeaways cite approximately $3.25. The analysis also highlights substantial financing risk, including the company's reported $4.1 million cash balance as of June 30, 2026.

The current FDA announcement supports one part of that thesis by providing greater clarity around a potential registrational pathway in colorectal cancer. However, it also reinforces the need for successful clinical data and adequate funding.

What investors may watch next

The most important near-term catalyst is the initial clinical data from REO 033 Part A.

Investors may also watch whether Oncolytics holds its End-of-Phase meeting with the FDA and reaches agreement on the final pivotal study design.

Beyond regulatory milestones, financing will remain important. The company intends to prepare for Part B while waiting for clinical data, making capital availability a key consideration if the program advances.

Ultimately, the market will be looking for evidence that the promising signals seen in earlier studies can translate into stronger randomized clinical results.

About the company

Oncolytics Biotech is a clinical-stage biotechnology company developing pelareorep, an intravenously delivered investigational immunotherapy designed to activate anti-tumor immune responses. The company is advancing pelareorep across gastrointestinal cancers, including colorectal, anal and pancreatic cancer.
